Your question deserves an answer but I'm not sure mine is adequate. In a heads up game I don't think you can and maybe shouldn't try put an opponent on a specific hand. IMO the best thing to do is assign probability estimates for possible hands. In a short handed game reading hands gets a little easier but is still problematic because often the contest for the pot is reduced to heads up play. However, I would think that in a 3 handed game you could infer somethings from how the button plays. With more players you can possibly draw inferences from other positions.
