You're only losing to AA, KK, JJ, and the case 6. The big pairs all seem pretty unlikely because you'd probably have seen a pre-flop raise. Loose passive players might slowplay a hand like AA here,...

This should be a standard three-bet against players in those positions, pretty much regardless of reads. The only time I might not raise this is from like a super-tight EP raiser or something.

On the flop, you really should wait until the turn. Raise any non-/images/graemlins/spade.gif, J, 9, or Q turn.
Consider folding if any of those cards hit.
