This was my second hand so I have no reads on opponent. What should my play be on the river and any comments on the flop and turn would be appreciated.

Bet 200. If you think it will look suspicious, bet 300. Hopefully button wasn't slow playing a set. If you don't think he'll bet the pot or close to it, check call that amount...

I would lead the flop. You want to get raised here. The c/r suggests that you have a strong hand, at least two pair out of the sb on a flop of rags and probably better. Leading out makes it harder to put you on a hand. You might even get raised by a flush draw.

The Villian's flat call on the turn is a bit scary, but if you get too scared when people call you can't play. He either has two pair (sixes and fours), made a boat or has some kind of a draw, most likely a combo draw, maybe 57 [img]/images/graemlins/spade.gif[/img] but I bet A2 [img]/images/graemlins/spade.gif[/img]

The 2 [img]/images/graemlins/club.gif[/img] is either irrelvant or helped you by counterfeiting him. Your perfect world would be if he had A5 [img]/images/graemlins/spade.gif[/img]

I think you have to bet out. $300 seems about right.

If his most likely holdings are a draw or a boat, isn't a river check call the best play? Or do you think he would pay off a $200 or $300 bet on a "combo" draw hand with some part of the board, hoping I missed my flush draw?

Funny how you think my flop CR here screams big hand, while in the other hand, someone else thought my flop CR screamed draw.

Results and Thoughts
 
I was really torn between betting out on the river or check calling and I am still not sure which is best. I decided to check call his $400 bet. He had 63s for the flopped 2-pair and turned full-house.
