I made my first couple trips to Foxwoods last week and I heard people yelling this. What does it mean?

Players saying it are indicating their desire to have a certain seat reserved for them.
Floorperson saying it is telling the dealer to reserve the seat for the new player.

If your initials are ASS, when they announce your name for a seat, Scream "lock it up for Ass". They will save the seat for you.

"lock it up" is also dealer jargon for taking losing bets and placing them in the dealer's tray.
