I have a little over 10k hands on PT. My comp has 512 RAM and runs [censored]' fast. Will I lose anything by compacting a database?

No.

Just note that even on a fast machine with any amount of memory, compacting will make PT appear to be Not Responding. Don't worry, just let it finish. This can also affect your processor usage, heating it up if you're overclocked much like a CPU intensive game and slowing down other apps. The only App that freezes on me when I am compacting though, other than PT, is Ho.exe, the Absolute table engine.

When it makes a backup to your database, you lose a little over the size of the original database in HD space.

Pokertracker uses Microsoft Access as its database engine. When you compact the database, it is basically taking out empty space that is in the database left over from the insertion, deletion, and merging of records. The backup .mdb file can be deleted without consequence once the compact operation successfully completes.
